Activation of protease‑activated receptor‑2 is associated with increased expression of inflammatory factors in the adipose tissues of obese mice.

چکیده
Previous studies have indicated that mast cells are critical for the pathogenesis of inflammatory diseases. Proteases released from mast cells have been reported to stimulate protease‑activated receptors (PAR), which induces microleakage and widespread inflammation. In order to investigate the pro‑inflammatory effect of PAR‑2 activation on adipose inflammation in obese mice, the varying distributions of macrophages and PAR‑2 in adipose tissue samples were compared between C57BL/6J (C57) and obese mice [B6(D)‑Leprdb/J, BKS(D)‑Leprdb/J and B6.V‑Lepob/J (ob/ob)] using immunohistochemical staining. Murine primary adipocytes and bone marrow‑derived macrophages (BMDMs) were used and the alterations in expression levels of inflammatory factors, induced by PAR‑2 activation, were detected by reverse transcription‑quantitative polymerase chain reaction and ELISA. In addition, the migratory capacity of primary BMDMs and the macrophage cell line, RAW264.7 were evaluated by co‑culture with primary adipocytes. The current study demonstrated a larger number of macrophages in the adipose tissues of obese mice compared with C57 mice. Furthermore, PAR‑2 expression was detected in various adipose tissues of mice and the protein expression levels of PAR‑2 were observed to be significantly higher in the total adipose tissues of ob/ob mice when compared with the C57 mice. The expression levels of inflammatory factors were increased in adipocytes and macrophages, and enhanced migratory ability was observed in macrophages pretreated with PAR‑2 agonists. The data of the current study suggests that PAR‑2 is involved in the process of obesity‑associated chronic low‑grade systemic inflammation, which indicates that the PAR‑2 signaling pathway may be a potential target for the treatment of obesity and its associated diseases.

The Association of Omentin Gene Expression in Visceral and Subcutaneous Adipose Tissues with Plasma Fatty Acids Profile and Dietary Fatty Acids
Introduction: Omentin, an adipokine, with anti-inflammatory effects reduces insulin resistance, and can hence, play an important role in prevention of cardiovascular disease and diabetes. The present study aimed to investigate the association of plasma and dietary fatty acids with gene expression of omentin in visceral and subcutaneous adipose tissues. Materials and Methods: Visceral and subcut...
متن کاملThe Association of Daily Physical Activity and Apelin Gene Expression and Serum Concentration in Omental and Subcutaneous Adipose Tissues of Obese and Morbid Obese Adults
Introduction: Adipose tissue affects body hemostasis by secreting a variety of proteins named adipokines. Plasma levels of apelin decreases after exercise; however, the evidence on gene regulation in adipocytes is rare. The aim of this study was to investigate the association of daily physical activity and applein gene expression and its serum levels in visceral and subcutaneous adipose tissues...

Objective(s): This study aims to investigate the pathogenicity and possible mechanisms of S100A9 function in mice models of scleroderma. Materials and Methods: The content of S100A9 in the skin tissues of mice with scleroderma was determined. Different concentrations of bleomycin (BLM) and S100A9 were subcutaneously injected into the backs of mice simultaneously, and then pathological changes i...
متن کاملEffect of blockade of neuropeptide Y receptor on aortic intima-media thickness and adipose tissue characteristics in normal and obese mice
Objective(s): Atherosclerosis is an important risk factor for coronary heart disease. Neuropeptide Y (NPY) and its receptors, located in peripheral tissue such as white adipose tissue, have been linked to obesity and fat storage. The role of NPY in atherosclerosis has not yet been fully studied, so this study was conducted to further investigate the effect of BIIE 0246, an NPY receptor antagoni...

Objective(s): Atherosclerosis is a chronic immune-inflammatory disease that generally leads to ischemic heart disease. Ghrelin has several modulatory effects on cardiovascular system. In this study, we investigated the effect of ghrelin on aortic intima-media thickness, size and the number of adipocyte cells in obese and control mice. Materials and Methods:This study was conducted on 24 male C...
